The Island Roads-led project, to upgrade lights and crossings at the busy Newport roundabout, paused over Christmas, but starts again today (Monday, January 10).
This morning's rush hour ran normally, but the road has now been shut to traffic.
It will affect motorists heading home from this evening, with a diversion along Quay Street.
The front doors to Newport police station and the home of Isle of Wight Council, County Hall, both sit on the closed stretch of road.
A number of on-street parking spaces have been suspended.
