Emergency crews were called to a crash near Ventnor, on Whitwell Road, at the junction with Week Lane and St Lawrence Shute.
The road is closed between St Lawrence Shute and Whitwell.
Cars are diverting via Wroxall and Godshill.
A partial road closure was already in place on Lower Gills Cliff Road, after an earlier landslip damaged traffic lights.
